Description

Tu Youyou found the lead in 1972 in a fourth-century Chinese manual that said to prepare the wormwood cold; heat destroyed the active principle. Printed here is the molecule with the bridge of two oxygens that makes it work.

Exploration

Inside the parasite there is free iron from the haemoglobin it has just digested. That iron breaks artemisinin's bridge of two oxygens and releases a radical that wrecks whatever is near it.

The drug is activated where the poison is. Outside the parasite the bridge does not break and the molecule does nothing.

It acts within hours, faster than any other antimalarial, and that is why it is used when the patient is already gravely ill.

Tu Youyou had hundreds of failed extracts behind her when she read in a fourth-century manual that wormwood is prepared by pressing it cold. Boiling it destroyed exactly that bridge.

It is never given alone. In monotherapy the parasite learns to go dormant for the hours it lasts, and wakes afterwards.
