Description

Ciclosporin turned up in a fungus from a soil sample from Hardangervidda and was approved in 1983; by not destroying the marrow, it made organ transplants possible. Printed here are its eleven amino acids in a ring, with no beginning and no end.

Exploration

Eleven amino acids closed in a ring, with no end to start from or to finish at, and several of them with the mirror form that normal proteins do not use. That is why no digestive enzyme knows how to attack it and it can be taken by mouth.

Inside the lymphocyte it binds to cyclophilin, and the resulting pair blocks calcineurin, which was the one about to give the order to make interleukin-2.

The T lymphocyte is neither destroyed nor exhausted: it is left without the signal to activate. The marrow stays intact and the patient keeps the rest of their defences.

That was what changed transplantation. Before, the whole immunity had to be levelled, and the recipient died of infection as often as of rejection.

It came from a fungus in a soil sample collected on the Hardangervidda plateau. It was approved in 1983 and one-year kidney graft survival went from around fifty per cent to more than eighty.
