Exploration

Folate is the piece the cell makes the bases of DNA with. Methotrexate resembles it so closely that it occupies the place in the DHFR enzyme, and holds on a thousand times more tightly than the original.

Without reduced folate there is no thymine, and without thymine there is no new DNA. The cell that was about to divide stops dead.

Farber first tried it the other way round, giving folate to children with leukaemia, and accelerated the disease. It was that acceleration that told him the opposite had to be done.

In 1948 he obtained the first remissions. They lasted weeks and the children died all the same, and even so it was the proof that a cancer could be pushed back with a chemical substance.

At doses a thousand times smaller it is used today against rheumatoid arthritis, where it does not act by that route but by releasing adenosine.
