Description

Paclitaxel was extracted from the bark of the Pacific yew, a tree that died when harvested, until it could be synthesised; it was approved in 1992. Printed here is the molecule that freezes tubulin instead of pulling it apart.

Exploration

Microtubules are tubes that are constantly assembled and dismantled, and during cell division they pull the chromosomes towards the two poles. The movement depends on their being able to come apart.

Paclitaxel sticks to already assembled tubulin and fixes it. The tube does not break, and a cell with a frozen spindle cannot share out its chromosomes or finish dividing.

Every earlier poison of this group stopped the tube being assembled. This one does the opposite and produces the same block.

It was extracted from the bark of the Pacific yew, a slow tree that died when harvested. A single treatment consumed several mature specimens.

Semi-synthesis from the leaves of the European yew, which grow back, solved the problem at the start of the nineties. Before that, supply was decided in a forest.
