Description

Published in 2019, prime editing carries with it the sequence it wants to write and copies it onto the DNA, instead of cutting and waiting for the cell to repair. Printed here is that direct writing.

CRISPR cuts and waits. Base editing converts a letter by chemistry. Prime editing does another thing: it brings with it the sequence it wants to install.

The guide not only indicates the site, it also carries the new text written at its end. A reverse transcriptase coupled to the enzyme copies that text directly onto one of the strands of the DNA.

There is no double-strand cut, no haphazard repair and no waiting for the cell to do anything. It is written over the top.

In theory it covers any substitution, and also small insertions and deletions, which adds up to the great majority of the known pathogenic mutations.

It was published in 2019 and there is still no approved therapy. The efficiency inside a living tissue and the size of the assembly, which does not fit comfortably into an AAV virus, are the two obstacles.
